Identification of Target Proteins for Promoting Nuclear Envelope Rupture in Cancer Cells

چکیده
The nuclear envelope separates and protects DNA from the cytoplasm. In cancer cells, frequent morphological abnormalities transient rupture of membrane promote damage malignancy. However, instability, acting as a tumor-promotion factor, can be viewed fatal weakness cells. Although exact mechanism is not clear, it known that loss tumor suppressor gene TP53 involved in instability this study, we demonstrate inhibition NUP93, which subunit pore complex, induces cell death by enhancing instability. RNA interference (RNAi) inhibiting NUP93 expression human line RPE1 caused rupture, damage, leading to death. Under condition was simultaneously suppressed with were significantly increased. Conversely, slightly decreased. decrease may ascribed fact induction apoptosis damage. We speculate amplification induce cell-specific synthetic lethality context TP53-independent Moreover, our results suggest proteins promising targets for development lethal anticancer drugs.

Nuclear envelope rupture drives genome instability in cancer
The nuclear envelope, composed of two lipid bilayers and numerous accessory proteins, has evolved to house the genetic material of all eukaryotic cells. In so doing, the nuclear envelope provides a physical barrier between chromosomes and the cytoplasm. Once believed to be highly stable, recent studies demonstrate that the nuclear envelope is prone to rupture. These rupture events expose chromo...
متن کاملDiffusion of proteins across the nuclear envelope of HeLa cells.
We describe an experimental system to study nucleocytoplasmic diffusion of proteins in living HeLa cells. To localize proteins to the nucleus, substrates were created that contain a nuclear localization sequence fused to Aequorea victoria green fluorescent protein (GFP). Transiently and stably transfected HeLa cells were used for these assays. A protein of 29-kDa molecular mass that harbors GFP...

conclusions this study provided conserved b-cell epitopes and peptides that can be useful for designing entry inhibitors and vaccines able to cover a global population, especially where genotype 5a is common. results differences in the probability of glycosylation in e1 and e2 regions were observed in this study. three conserved antigenic b-cell epitopes were predicted in the e2 regions and als...
متن کاملStudy of NGEP expression in androgen sensitive prostate cancer cells: A potential target for immunotherapy
Background: Prostate cancer is one of the leading causes of cancer deaths among men. New gene expressed in prostate (NGEP), is a prostate-specific gene expressed only in normal prostate and prostate cancer tissue. Because of its selective expression in prostate cancer cell surface, NGEP is a potential immunotherapeutic target. To target the NGEP in prostate cancer, it is essential to investig...
متن کاملW4: Cancer Stem Cells as a New Target Point for Treatment of Glioblastoma
Glioblastoma is a destructive form of brain tumor that kills most patients within two years of diagnosis.Treatments for glioblastoma include the usual options of surgery, radiation therapy and chemotherapy, but there is no effective treatment. The tumors are capable of spreading tendrils out into the brain and it can grow back in a matter of months after being removed. The cancer stem cell...
